B cell-intrinsic mechanisms in the pathogenesis of Systemic Lupus Erythematosus (SLE)

Research Summary

The Jackson Laboratory uses both murine models and human samples to interrogate the function of B cells in systemic lupus erythematosus (SLE). For example, we have shown that B cell-intrinsic Toll-like receptor (TLR) signals, B cell antigen presentation to cognate CD4+ T cells, B cell-derived co-stimulation, and cell-intrinsic cytokine signals each coordinate to orchestrate the formation of spontaneous, autoimmune germinal centers (GC). Together, these studies have lent support to an emerging paradigm of humoral autoimmunity wherein B cells orchestrate initial breaks in immune tolerance.
